Interfacial layering in a three-component polymer system
We study theoretically the temporal evolution and the spatial structure of
the interface between two polymer melts involving three different species (A,
A* and B). The first melt is composed of two different polymer species A and A*
which are fairly indifferent to one another (Flory parameter chi_AA* ~ 0). The
second melt is made of a pure polymer B which is strongly attracted to species
A (chi_AB 0). We then show
that, due to these contradictory tendencies, interesting properties arise
during the evolution of the interface after the melts are put into contact: as
diffusion proceeds, the interface structures into several adjacent
"compartments", or layers, of differing chemical compositions, and in addition,
the central mixing layer grows in a very asymmetric fashion. Such unusual
behaviour might lead to interesting mechanical properties, and demonstrates on
a specific case the potential richness of multi-component polymer interfaces
(as compared to conventional two-component interfaces) for various
applications.Comment: Revised version, to appear in Macromolecule
The composition of the protosolar disk and the formation conditions for comets
Conditions in the protosolar nebula have left their mark in the composition
of cometary volatiles, thought to be some of the most pristine material in the
solar system. Cometary compositions represent the end point of processing that
began in the parent molecular cloud core and continued through the collapse of
that core to form the protosun and the solar nebula, and finally during the
evolution of the solar nebula itself as the cometary bodies were accreting.
Disentangling the effects of the various epochs on the final composition of a
comet is complicated. But comets are not the only source of information about
the solar nebula. Protostellar disks around young stars similar to the protosun
provide a way of investigating the evolution of disks similar to the solar
nebula while they are in the process of evolving to form their own solar
systems. In this way we can learn about the physical and chemical conditions
under which comets formed, and about the types of dynamical processing that
shaped the solar system we see today.
This paper summarizes some recent contributions to our understanding of both
cometary volatiles and the composition, structure and evolution of protostellar
disks.Comment: To appear in Space Science Reviews. The final publication is

Molecular basis of antibiotic multiresistance transfer in Staphylococcus aureus
Multidrug-resistant Staphylococcus aureus infections pose a significant threat to human health. Antibiotic resistance is most commonly propagated by conjugative plasmids like pLW1043, the first vancomycin-resistant S. aureus vector identified in humans. We present the molecular basis for resistance transmission by the nicking enzyme in S. aureus (NES), which is essential for conjugative transfer. NES initiates and terminates the transfer of plasmids that variously confer resistance to a range of drugs, including vancomycin, gentamicin, and mupirocin. The NES N-terminal relaxase–DNA complex crystal structure reveals unique protein–DNA contacts essential in vitro and for conjugation in S. aureus. Using this structural information, we designed a DNA minor groove-targeted polyamide that inhibits NES with low micromolar efficacy. The crystal structure of the 341-residue C-terminal region outlines a unique architecture; in vitro and cell-based studies further establish that it is essential for conjugation and regulates the activity of the N-terminal relaxase. This conclusion is supported by a small-angle X-ray scattering structure of a full-length, 665-residue NES–DNA complex. Together, these data reveal the structural basis for antibiotic multiresistance acquisition by S. aureus and suggest novel strategies for therapeutic intervention
Mechanism of subunit interaction at ketosynthase-dehydratase junctions in trans-AT polyketide synthases
Modular polyketide synthases (PKSs) produce numerous structurally complex natural products with diverse applications in medicine and agriculture. They typically consist of several multienzyme subunits that utilize structurally-defined docking domains (DDs) at their N- and C-termini to ensure correct assembly into functional multi-protein complexes. Here we report a fundamentally different mechanism for subunit assembly in trans-AT modular PKSs at the junction between ketosynthase (KS) and dehydratase (DH) domains. This involves direct interaction of a largely unstructured docking domain (DD) at the C-terminus of the KS with the surface of the downstream DH. Acyl transfer assays and mechanism-based cross-linking established that the DD is required for the KS to communicate with the acyl carrier protein appended to the DH. Two distinct regions for binding of the DD to the DH were identified using NMR spectroscopy, carbene foot-printing and mutagenesis, providing a foundation for future elucidation of the molecular basis for interaction specificity
Search for the standard model Higgs boson in the H to ZZ to 2l 2nu channel in pp collisions at sqrt(s) = 7 TeV
A search for the standard model Higgs boson in the H to ZZ to 2l 2nu decay
channel, where l = e or mu, in pp collisions at a center-of-mass energy of 7
TeV is presented. The data were collected at the LHC, with the CMS detector,
and correspond to an integrated luminosity of 4.6 inverse femtobarns. No
significant excess is observed above the background expectation, and upper
limits are set on the Higgs boson production cross section. The presence of the
standard model Higgs boson with a mass in the 270-440 GeV range is excluded at
95% confidence level.Comment: Submitted to JHE
Combined search for the quarks of a sequential fourth generation
Results are presented from a search for a fourth generation of quarks
produced singly or in pairs in a data set corresponding to an integrated
luminosity of 5 inverse femtobarns recorded by the CMS experiment at the LHC in
2011. A novel strategy has been developed for a combined search for quarks of
the up and down type in decay channels with at least one isolated muon or
electron. Limits on the mass of the fourth-generation quarks and the relevant
Cabibbo-Kobayashi-Maskawa matrix elements are derived in the context of a
simple extension of the standard model with a sequential fourth generation of
fermions. The existence of mass-degenerate fourth-generation quarks with masses
below 685 GeV is excluded at 95% confidence level for minimal off-diagonal
mixing between the third- and the fourth-generation quarks. With a mass
difference of 25 GeV between the quark masses, the obtained limit on the masses
of the fourth-generation quarks shifts by about +/- 20 GeV. These results
significantly reduce the allowed parameter space for a fourth generation of
fermions.Comment: Replaced with published version. Added journal reference and DO
Measurement of the t t-bar production cross section in the dilepton channel in pp collisions at sqrt(s) = 7 TeV
The t t-bar production cross section (sigma[t t-bar]) is measured in
proton-proton collisions at sqrt(s) = 7 TeV in data collected by the CMS
experiment, corresponding to an integrated luminosity of 2.3 inverse
femtobarns. The measurement is performed in events with two leptons (electrons
or muons) in the final state, at least two jets identified as jets originating
from b quarks, and the presence of an imbalance in transverse momentum. The
measured value of sigma[t t-bar] for a top-quark mass of 172.5 GeV is 161.9 +/-
2.5 (stat.) +5.1/-5.0 (syst.) +/- 3.6(lumi.) pb, consistent with the prediction
of the standard model.Comment: Replaced with published version. Included journal reference and DO
